第一、神經元過度興奮所致異常放電：用細胞內電極對癲癇動物模型大腦皮質過度興奮的描記過程發現，專家提醒，在神經元動作電位點燃暴發后，出現連續的去極化和超極化，產生興奮性突觸后電位(EPSP)和去極化飄移(DS)，使細胞內Ca2+和Na+增加，細胞外K+增加，Ca2+減少，出現大量DS，并以比正常傳導快數倍的速度向周圍神經元擴散。

第二、抑制性氨基酸(IAA)含量減少和GABA受體功能異常：癇性病灶中細胞外K+增加可減少IAA的釋放，降低具有突觸前抑制功能的GABA受體活動，使興奮性放電易于向周圍和遠隔區投射。

第三、興奮性氨基酸(EAA)過度釋放和NMDA受體活化：通過對癇性病灶的生物化學研究發現，海馬和顳葉神經元在去極化時可釋放大量EAA和其他神經遞質，激活NMDA受體后，大量的Ca2+內流，導致興奮性突觸的活動進一步增強。
