第二、抑制性氨基酸(IAA)含量減少和GABA受體功能異常：癇性病灶中細胞外K+增加可減少IAA的釋放，降低具有突觸前抑制功能的GABA受體活動，使興奮性放電易于向周圍和遠隔區投射。

第三、興奮性氨基酸(EAA)過度釋放和NMDA受體活化：通過對癇性病灶的生物化學研究發現，海馬和顳葉神經元在去極化時可釋放大量EAA和其他神經遞質，激活NMDA受體后，大量的Ca2+內流，導致興奮性突觸的活動進一步增強。
